چکیده

an inositol ring. Diphosphoinositol pentakisphosphate (IP7), the most abundant inositol pyrophosphate, is synthesized on phosphorylation of inositol hexakisphosphate (IP6) by IP6 kinases, of which there are 3 mammalian isoforms (IP6K1/2/3) and a single yeast isoform. Yeast lacking IP6 kinase are devoid of polyP, suggesting a role for IP6 kinase in maintaining polyP levels. We theorized that the molecular link between IP6 kinase and polyP is conserved in mammals and investigated whether polyP-dependent platelet

Deficiency of Src homology 2 domain-containing inositol 5-phosphatase 1 affects platelet responses and thrombus growth.

Platelets are critical for normal hemostasis. Their deregulation can lead to bleeding or to arterial thrombosis, a primary cause of heart attack and ischemic stroke. Src homology 2 domain-containing inositol 5-phosphatase 1 (SHIP1) is a 5-phosphatase capable of dephosphorylating the phosphatidylinositol 3,4,5-trisphosphate second messenger into phosphatidylinositol 3,4-bisphosphate. SHIP1 plays...

Intelligent platelet inhibitors are on the horizon.

Hemostasis is a tightly regulated physiological sequence of events that maintains vascular integrity and imminent blood loss after vessel injury. Platelets play a critical role in primary hemostasis and in regulation of coagulation. Under physiological conditions, hemostasis and thrombosis are in a fragile equilibrium. Uncontrolled platelet activation leads to intravascular thrombosis and organ...

Disabled-2 is required for efficient hemostasis and platelet activation by thrombin in mice.

OBJECTIVE The essential role of platelet activation in hemostasis and thrombotic diseases focuses attention on unveiling the underlying intracellular signals of platelet activation. Disabled-2 (Dab2) has been implicated in platelet aggregation and in the control of clotting responses. However, there is not yet any in vivo study to provide direct evidence for the role of Dab2 in hemostasis and p...

HEMOSTASIS, THROMBOSIS, AND VASCULAR BIOLOGY SHIP-2 forms a tetrameric complex with filamin, actin, and GPIb-IX-V: localization of SHIP-2 to the activated platelet actin cytoskeleton

The platelet receptor for the von Willebrand factor (VWF) glycoprotein Ib-IX-V (GPIb-IX-V) complex mediates platelet adhesion at sites of vascular injury. The cytoplasmic tail of the GPIb subunit interacts with the actin-binding protein, filamin, anchoring the receptor in the cytoskeleton. In motile cells, the second messenger phosphatidylinositol 3,4,5 trisphosphate (PtdIns(3,4,5)P3) induces s...

While not essential for normal thrombosis and hemostasis, platelet- derived VWF fosters ischemic stroke injury in mice

Von Willebrand factor (VWF) is a key hemostatic protein synthesized in both endothelial cells and megakaryocytes. Megakaryocyte-derived VWF is stored in αgranules of platelets and is enriched in hyperactive ‘ultra-large’ VWF multimers. To elucidate the specific contribution of platelet VWF in hemostasis and thrombosis we performed crossed bone marrow transplantations between C57BL/6J and Vwf mi...
